P21 (also referred to as P021 in academic papers) is a synthetic hexapeptide with sequence Ac-DGGLAG-NH₂, rationally derived from the biologically active domain of Ciliary Neurotrophic Factor (CNTF) by the New York State Institute for Basic Research. Native CNTF is limited by large molecular weight, poor blood-brain barrier penetration and obvious adverse reactions; terminal acetylation and amidation modification greatly enhance P21’s enzymatic hydrolysis resistance and CNS permeability, retaining neurotrophic effects while eliminating side effects of parent CNTF. All relevant data are limited to preclinical animal studies with no human clinical trials conducted. It is supplied as white lyophilized powder with HPLC purity ≥99%.

Main laboratory research applications:

1. Hippocampal neurogenesis & cognitive enhancement: Upregulate endogenous BDNF, boost newborn neuron proliferation and synaptic spine density in hippocampus, widely adopted in behavioral tests for learning and memory on Alzheimer’s and aged animal models.
2. Pathological protein clearance: Inhibit GSK-3β to reduce hyperphosphorylated Tau tangles and amyloid-beta aggregation, suitable for mechanism research on Alzheimer’s disease and tauopathies.
3. Neuroprotection against brain injury: Suppress NF-κB mediated neuroinflammation, scavenge reactive oxygen species, stabilize mitochondrial membrane and inhibit neuronal apoptosis for traumatic brain injury and cerebral ischemia researches.
4. Brain senescence & neurodegeneration exploration: Applied in studies of age-related cognitive decline and progressive neuronal degeneration, commonly used as control peptide alongside other nootropic research compounds.
